目的:研究喘平提取液(麻黄、洋金花)中麻黄碱及伪麻黄碱的挥发动力学。方法:将不同pH(2、4和6)的喘平提取液置于不同的温度(50、70、90℃)的水浴中,定时取样,HPLC检测溶液中麻黄碱与伪麻黄碱的残留浓度。根据一级动力学方程计算麻黄碱与伪麻黄碱的挥发动力学常数,根据阿仑尼乌斯公式计算不同pH值条件下,麻黄碱与伪麻黄碱的挥发活化能(Ea)。结果:在pH值2,4和6条件下,麻黄碱与伪麻黄碱的挥发活化能分别为30.204 kJ/mol和18.193 kJ/mol;24.896 kJ/mol和15.384 kJ/mol;22.206 kJ/mol和14.279 kJ/mol。麻黄碱与伪麻黄碱的挥发量与喘平溶液的蒸发量之间成线性相关。结论:麻黄碱与伪麻黄碱的挥发动力学研究为含麻黄单、复方提取浓缩及制剂工艺研究提供参考。
AIM:To study volatilization kinetics of ephedrine and pseudoephedrine in Chuanping Extract(Ephedre herba,Daturae flos).METHODS:Chuanping fluid extract at different pH(2,4 and 6) was placed in water bath at different temperature(50℃,70℃ and 90℃).The residual concentrations of ephedrine and pseudo-e-phedrine were determined by HPLC.The volatile kinetic constant of ephedrine and pseudoephedrine was calculated using first order kinetics equation.The volatile activation energy of ephedrine and pseudoephedrine was calculated using Arrhenius equation.RESULTS:Volatile activation energy of ephedrine and pseudo-ephedrine in Chuanping fluid extract was 30.204 kJ/mol and 18.193 kJ/mol at pH2;24.896 kJ/mol and 15.384 kJ/mol at pH4;22.206 kJ/mol and 14.279 kJ/mol at pH6 respectively.There existed a linear relationship between the volatilization amount of ephedrine and pseudoephedrine and the evaporation amount of Chuanping fluid extract.CONCLU-SION:This study on volatilization kinetics of ephedrine and pseudoephedrine can be applied to optimizing the ab-straction,concentration and preparation of single herbal and compound prescription containing ephedra.
