通过分子荧光、KI试验,证明了芘分子与DNA分子之间的结合方式是嵌插结合.而且通过引入环糊精(CD),考察了芘与DNA分子之间的结合方式.基于荧光光谱的改变,可以得出芘以及芘-CD被DNA猝灭的猝灭常数Ksv值以及与DNA相作用的结合常数(K)和结合位点数(n).实验结果证明,芘分子与DNA分子率之间主要是嵌插结合作用.
The interactive model of pyrene with DNA was investigated by means of the inclusion action of β- cyclodextrin. Through the changes of fluorescence spectra, the intrinsic binding constant (K) of pyrene with DNA and inclusion complex with DNA is obtained. In the case of Tris buffer solution (pH 7.2) ,their values are 3. 37 × 10^4 (mol/L)-1, and 3.04 × 10^4 (mol/L)-1, respectively. According to the experimental results,it can be inferred that the interactive model of pyrene with DNA is "intercalative binding".
