以水产养殖中鱼类致病细菌-嗜水气单胞菌为材料,采用DS法对该菌的低分子质量蛋白(LMW)进行分离提取,LMW蛋白的纯度为90%,通过LC MS/MS分析鉴定了97个蛋白,其中理论分子质量小于25 ku的蛋白有57个,这些蛋白在蛋白合成转录、细胞代谢调控等方面都具有重要功能.实验结果表明,该方法在研究LMW蛋白中具有广阔的应用潜力.
A differential solubilization extracting method called DS method was optimized to obtain low molecular weight( LMW)proteins from Aeromonas hydrophila,an aquatic pathogenic bacterium and above 90% purity of LMW proteins were acquired. 97 of them were successfully identified by LC MS / MS in these extracted proteins and 57 of them had the theoretical MWs lower than 25 ku. GO analysis revealed that these proteins involved in many biological processes such as biosynthesis,cellular metabolism and translation process. This extraction method has been proved to be effective for LMW proteins extraction.
