目的:观察神经干细胞(NSC)与原浆型星形胶质细胞(PAS)联合移植对大鼠损伤脊髓轴浆运输功能的修复作用。方法:40只Wistar大鼠,制成L1~L2左侧脊髓半切空洞损伤模型,随机分为损伤组(A组),损伤后移植PAS组(B组)、移植NSC组(C组)、NSC和PAS按2:1比例联合移植组(D组),每组10只。4周及8周后行损伤部位神经丝-200(NF-200)染色观察NSC在体内的分化情况,腓肠肌胆碱酯酶染色观察运动终板的反应和核黄逆行示踪观察轴浆运输的恢复情况。结果:(1)4周时D组和C组移植部位可见少量NF-200阳性标记的神经元.8周时数量明显增多,D组多于C组;4周及8周时A、B组均未见到明显阳性标记的神经元。(2)4周时.A、B组胆碱酯酶染色示运动终板均出现退变,终板染色变浅;8周时A组终板明显退变,出现大片染色空白区,甚至终板消失,只剩下模糊的轮廓;B组终板退变程度轻于A组,着色淡,轮廓欠清晰,周边呈浅棕红色淡染,但未出现大片染色空白区;C、D组4周时终板边缘发生皱缩,呈颗粒样改变,无明显染色缺失;8周时C组较4周时变化不明显,D组受损终板皱缩明显好转,颗粒样改变消失。(3)4周时,B、C、D组核黄染色的神经元散在位于脊髓的前、后、侧角,其中D组阳性标记的神经元的数量及荧光强度大于B、C组,C组又稍强于B;8周时B、C、D组阳性神经元数量均增多.神经元的数量及荧光强度仍是D组〉C组〉B组;A组4周及8周时均未见到明显阳性染色的神经元。结论:联合移植NSC和PAS(2:1)能更好地修复损伤脊髓的轴浆运输功能并能较好地防治其后肢肌肉运动终板的溃变。
Objective:To observe the effects of neural stem cells(NSC) and protoplasmic astrocyte(PAS) on the repair of axon-plasma transporting of injured spinal cord in adult rats.Method:Forty Wistar rats were respectively divided into four groups:hemisection-cavity injured group (group A),PAS-transplant group(group B),NSC-transplant group(group C),NSC and PAS in the ratio of 2:1 transplant group(group D).NF-200 immunohistochemical staining was carried out at the 4th and 8th week after operation to observe the differentiation of NSC in vivo,at the same time AchE staining and NY tracing were carried out to observe the recovery of axon-plasma transporting and the protective effects of motor end-plate.Reset: (1)With NF-200 staining,at 4 weeks there were a few neurons in the group D and C,at 8 weeks the number of neurons increased obviously in both two groups,but the number in the group D was more than that in group C.At 4 and 8 weeks there was not any positive neuron found in group A and B.(2)With AchE staining,at 4 weeks the morphology of motor end-plate degenerated in group A and B,at 8 weeks the morphology of motor end-plate degenerated markedly or even disappeared in group A,but the degenerative reaction in group B was mild compared with group A.At 4 weeks,particles and crimple of edge were observed on motor end-plate in group C and D. At 8 weeks,there was no remarkble change in group C compared with that at 4 weeks,the morphology of motor end-plate was nearly normal in group D.(3)With NY tracing,at 4 weeks neurons were interspered in anterior and posterior horns in group B,C,D.The positive neurons in group D was much more than that in group B and C,and group C was more than group B.At 8 weeks the number of positive neurons increased obviously among the three groups,and the change of the number and fluorescence intensity of positive neurons was D〉C〉B in the three groups,in group A,there was no obviously positive neuron at 4 and 8 weeks.
