目的通过实施冠状动脉内皮损伤术,加速小型猪在高胆固醇饮食条件下的动脉粥样硬化进程,建立小型猪定位冠状动脉硬化技术,为心血管疾病研究提供理想的动物模型。方法采用8头五指山小型猪进行冠状动脉内皮损伤术,术前饲喂3.0%高胆固醇饲料20—25d,在冠状动脉前降支中段进行球囊扩张,压力为6—10atm,扩张次数3次。结果除1头小型猪,在经过扩张压力10个大气压,扩张3次手术后存活外,其余小型猪在术后6—24h内死亡。结论五指山小型猪冠状动脉扩张后,极易出现室颤,推测该品种小型猪对这种手术的耐受力较差。
Objective Coronary balloon angioplasty may accelerate atherosclerosis procedure in atherogenic diet fed pigs. The aim of this study was to explore the possibility of establishment of a localized atherosclerosis model in Wuzhishan minipigs and provide suitable animal model for studying cardiovascular diseases. Methods Coronary balloon angioplasty was performed in 8 Wuzhishan mini-pigs, which had fed on atherogenic diet containing 3.0% cholesterol for 20 - 25 days. Coronary balloon angioplasty was conducted at the middle and upper segments of the left anterior descending arteries. Angioplasty was performed by three inflations, using a pressure of 6 - 10 atmospheres. Each inflation lasted 10 s, with an interval of 60 s between inflations. Results One miniature pig survived after three inflations, which received 10 atmospheres inflation. Other miniature pigs died within 6 - 24 hours after successful inflation. The course of death was ventricular fibrillation. Conclusion To our knowledge this is the first report for conducting coronary balloon angioplasty in hypedipidemic Wuzhishan mini-pigs. The results of our study indicate that Wuzhishan miniature pigs poorly stand coronary balloon angioplasty and are prone to occur ventricular fibrillation after this surgery. Therefore, we would believe that Wuzhishan miniature pigs might be not suitable for this type of studies.
