从云南鸢尾上采集到疑似感染番茄斑萎病毒属病毒的样本KBG-IR,经过TSWV和INSV免疫试纸条检测、RT-PCR,克隆和测序.获得病毒样本NP (834 bp),NSs(1380 bp),NSm (930 bp)以及部分L RNA片段(923 bp).测序分析表明,KBG-IR分离物所扩增片段与番茄环纹斑点病毒(TZSV)的核苷酸序列同源性达到96.1% ~97.8%,氨基酸序列同源性为96.1% ~ 100%.构建系统进化树也体现了序列的高度一致性.寄主范围测定结果与TZSV相近.这些数据表明该病毒为番茄环纹斑点病毒,这是首次在鸢尾上发现TZSV.
The sample (KBG-IR) was suspected to be infected by tospovirus, which was collected from Iris in Yunnan province. It was tested by TSWV and INSV immunostrip assay test, reverse tran- scription (RT) -PCR, clone and sequencing. The complete NP, NSs, NSm and partial L RNA se- quences of the KBG-IR isolate were 834, 1380, 930 and 923bp in length, respectively. Sequence a- nalysis showed high nucleotide homology (96. 1% -97.8% ) and amino acid sequences homology (96. 1% -100% ) to that of Tomato zonate spot virus (TZSV). Phylogenetic analysis revealed that this isolate clustered with TZSV, and host range test also indicated it was closest relation to TZSV. These data suggested the pathogen was TZSV, and this is the first report TZSV infected on Iris.
