目的观察CD80,CD86,Foxp3,Galectin-9,PD-L1在小鼠DC细胞的表达,了解小鼠DC2.4细胞系的免疫学特性。方法用含10%胎牛血清的RPMI-1640培养液培养小鼠DC2.4细胞,光学显微镜观察细胞形态;流式细胞术检测CD80,CD86,Foxp3,Galectin-9,PD-L1在DC2.4细胞表达。结果光学显微镜示小鼠DC2.4细胞贴壁生长,呈树突、梭形及不规则形;流式细胞术检测DC2.4细胞CD80表达为91.04%±4.90%,CD86表达为98.54%±1.96%,Foxp3无或微表达,Galectin-9表达为14.33%±2.94%,PD-L1表达为98.80%±0.63%。结论小鼠DC2.4细胞是一个高表达CD80,CD86及PD-L1,低表达Galectin-9,无或微表达Foxp3的细胞系。

Objective To observe the expression of CD80,CD86,Foxp3,Galectin-9 and PD-L1 in mouse dendritic cells(DC) and to study the immunological characteristic of mouse DC2.4 cell line.Methods Morphology of mouse DC2.4 cells was observed under an optical microscope after the cells were cultured with RPMI-1640 culture medium containing 10% fetal calf serum(FCS).Expressions of Foxp3,Galectin-9 and PD-L1 in DC cells were detected by flow cytometry(FCM).Results Optical microscopy revealed that the DC2.4 cells grew along the wall of DC and were dendritic,spindle,and irregular in shape.Flow cytometry showed that 91.04%±4.90% of CD80 and 98.54%±1.96% of CD86 were expressed in DC2.4 cells,respectively.Foxp3 was not or seldom expressed,while 14.33%±2.94% of Galectin-9 and 98.80%±0.63% of PD-L1 were expressed,respectively,in DC2.4 cells.Conclusion CD80,CD86 and PD-L1 are highly expressed,Galectin-9 is slowly expressed,and Foxp3 is not or seldom expressed in DC2.4 cell line.
