目的研究ERK-1/2对N/OFQ调节大鼠皮层神经元延迟整流钾电流(Iκ)激活动力学的影响,初步探讨N/OFQ调节大鼠皮层神经元Iκ的ERK.1/2途径。方法研究采用全细胞膜片钳技术,观察ERK-1/2特异性抑制剂U0126对N/OFQ调节大鼠皮层神经元Iκ激活动力学的影响。结果在ERK-1/2抑制剂U0126存在情况下,IK激活曲线的V1/2由给药前的(-42±3.7)mV向超极化方向移动为(-39.9±5.9)mV(P〈0.01)。Iκ激活曲线的K由给药前的(21.6±8.1)mV变为给药后的(22.9±3.8)mV(P〈0.05)。结论U0126可以显著抑制N/OFQ对Iκ激活曲线的影响。
Objective To investigate the effects of ERK-1/2 on modulation of delayed rectifier potassium current ( Iκ ) activation kinesics by N/OFQ in acutely dissociated rat cerebral parietal cortical neurons. Method Whole-cell voltage clamp technique was used. Result In presence of U0126, inhibitor of ERK-1/2, 0.1μmol/L N/OFQ shifted the activation curve of Iκ to hyperpolarize potential from ( -42 ± 3.7) mV to ( -39.9 ± 5.9) mV (P 〈0.01 ) and changed the slope factor (K) of the activation curve from (21.6 ± 8.1) mV to (22.9 ± 3.8) mV (P〈0.05). Conclusion These data sustained our contention that U0126 inhibited the positive shift of Iκ activation induced by N/OFQ.
