目的:筛选磁性葡聚糖一丝裂霉素前体药物的制备工艺,井初步考察其性质。方法:利用正交试验设计考察了制备所需的pH值、药物与栽体葡聚糖的重量比、药物与催化荆的重量比以及反应浓度等对接枝率、栽药量以及粒径的影响。结果:在pH5.0~5.5时。药物与葡聚糖重量比为1.5:10.1-乙基-3-(3-二甲基氨丙基)-碳二亚胺(EDC)与葡聚糖重量比为2:1的条件下,所得到的接枝率可达到91.7%,栽药量达到8.57%,粒径平均为135nm,跨距小于0.30。结论:通过正交设计得到了高接枝率和栽药量的磁性前体纳米药物。
ORJECTIVE To optimize the preparation technology of the prodrug magnetic dextran-mitomycin nanoparticles. METHODS The effects of the factors were reviewed by the orthogonal-test method including the reaction pH, the scale of the reactant and the concentration of the solution. RESULTS The highest grafting coefficient and loading capacity were occurred at the pH 5.0-5.5, the ratios of the mltomycin and catalyzer to the dextran were 1.5:10 and 2:1 respectively by weight. CONCLUSION The diameter of the nanoparticles of the result ant product was about 49 nm, and a high grafting coefficient reached to 91.7% and loading capacity reached to 8.57% respectively.
