VASA样蛋白是决定生殖系发育的重要调控蛋白因子之一,具有广泛的保守性,因此vasa的表达被认为是生殖细胞中最可靠的分子标记。为研究家蚕vasa样基因(Bombyx mori vasa-like gene,Bmvlg)的基本结构特征,通过RT-PCR从家蚕雄性性腺组织中获得了4个不同长度的BmvlgcDNA片段。经克隆分析,其中一个完整的BmvlgcD-NA全长1 806 bp,编码全长601个氨基酸的家蚕VASA样蛋白,该蛋白的结构特征与已知同源VASA样蛋白的结构特征基本一致;另3个不同长度的BmvlgcDNA均有各自的起始密码子和终止密码子。推测Bmvlg基因可能存在着多种可变剪接。
VASA-like protein is one of the key regulatory factors in determining development of germ line in Drosophila. It is conserved across species. Therefore, the expression of vasa is considered to be one of the most reliable molecular indications of germ cells. In order to study the Bombyx mori vasa-like gene ( Bmvlg), four different-length fragments from Bombyx mori 5th instar larval testis were obtained by RT-PCR ( reverse transcription and polymerase chain reaction). Further cloning experiments showed that the full-length Bmvlg cDNA was 1 806 nucleotide long and encoded a 601 amino acid protein ( Bmvasa-like protein), which contained almost all of the known characteristics of vasa homologs. The other three fragments of different lengths were found to contain their own start and stop codons, respectively, suggesting that Bmvlg gene products have several alternatively spliced forms.
