目的 评价利奈唑胺和磷霉素联用对临床分离60株甲氧西林耐药的金黄色葡萄球菌(MRSA)的体外抗菌活性。方法 用棋盘法设计,微量肉汤稀释法测定不同浓度组合的抗菌药物对60株临床分离MRSA最低抑菌浓度,并计算部分抑菌浓度(FIC)指数判定联合效应。测定利奈唑胺与磷霉素联用或单用对2株MRSA临床分离菌的体外杀菌曲线。结果 利奈唑胺与磷霉素联合用药后对60株MRSA临床分菌株的MIC50、MIC90较单药组降低。利奈唑胺与磷霉素联合应用对60株MRSA以协同作用为主(71.67%)。联合用药的杀菌曲线的细菌数量较单药相比降低〉2 log10CFU·m L-1,显示协同效应。结论 利奈唑胺与磷霉素联合用药在体外对MRSA以协同作用为主,杀菌曲线也显示联合用药有协同效应。
Objective To evaluate the antibiotic effects of linezolid combined with fosfomycin against 60 strains of methicillin - resistant Staphylococcus aureus (MRSA) in vitro. Methods The broth microdilu- tion and checkerboard method were used to determine the MICs of linezolid or fosfomycin against 60 strains of MRSA and fractional inhibito- ry concentration (FIC) index was calculated according to MIC results. The time - killing curves of linezolid and fosfomycin alone or combination were determined. Results Of the tested strains, 43 isolates (71.67% ) showed synergistic interaction (FICI≤0. 5) was observed. At 24 h the combination resulted in more than 2 log10 CFU mL-l reduction in viable bacteria compared with linezolid or fosfomycin alone. Conclusion The results suggested high synergy with linezolid and fosfomycin combination against MRSA strains.
