目的观察同型半胱氨酸(Hcy)对人血管平滑肌细胞(VSMCs)增殖及凋亡的影响.方法采用组织贴块法体外培养人VSMCs,将不同浓度Hcy与VSMCs孵育不同时间,应用MTT法检测VSMCs细胞增殖、流式细胞术(FCM)检测VSMCs细胞凋亡.结果体外培养的人VSMCs在终浓度为500 μmol/L Hcy的培养液中孵育0 h、6 h、12 h、24h、48 h后的A值分别为0.504±0.053、0.586±0.043、0.625±0.031、0.683±0.087、0.812±0.147,细胞凋亡率分别为2.39%±0.47%、2.45%±0.96%、7.58%±2.02%、13.37%±4.71%、17.69%±3.13%,表明Hcy呈时间依赖性诱导人VSMCs细胞增殖和细胞凋亡;在终浓度分别为0、100、200、500、1 000 μmol/L Hcy的培养液中孵育24 h后的A值分别为0.549±0.067、0.616±0.121、0.644±0.095、0.685±0.139、0.846±0.142,细胞凋亡率分别为2.68%±0.23%、2.79%±0.89%、8.45%±2.41%、13.41%±4.98%、17.75%±5.56%,表明Hcy呈浓度依赖性诱导人VSMCs细胞增殖和细胞凋亡.结论Hcy可以诱导VSMCs增殖和细胞凋亡,这一双重作用可能是Hcy致动脉粥样硬化作用的机制之一.
Objective To investigate the effects of Homocysteine(Hey) on the proliferation and apoptosis of human vascular smooth muscle cells(VSMCs) cultured in vitro. Methods VSMCs were cultured in vitro and were incubated with various dosage Hcy for various time. The rate of proliferation of VSMCs was detected by the way of MTT. The rate of apoptosis of VSMCs was detected by the way of flow cytometry. Results The absorbency of MTT of VSMCs incubated with 500 μmol/L Hcy for 0 h ,6 h ,12 h ,24 h ,48 h was 0. 504 ±0. 053,0.586 ±0. 043,0. 625 ±0. 031,0. 683 ±0. 087,0. 812 ±0. 147, respectively,the index of apoptosis was 2.39% ± 0.47 % ,2.45 % 0.96% ,7.58 % ± 2.02%, 13.37 % ± 4.71%, 17.69 % ± 3.13 %, respectively, revealing that Hcy could induce proliferation and apoptosis of VSMCs in a time-dependent manner. The absorbency of MTT of VSMCs incubated with 0,100,200,500,1000 μmol/L Hey for 24 h was 0. 549 ± 0. 067,0. 616 ±0. 121,0. 644 ± 0. 095,0. 685 ± 0. 139,0. 846 ± 0. 142, respectively, the index of apoptosis was 2.68% ± 0.23% ,2.79% ± 0.89% ,8.45% ± 2.41%, 13.41% ± 4.98%, 17.75% ± 5.56%, respectively, revealing that Hey could induce the proliferation and apoptosis of VSMCs in a dose-dependent manner. Conclusion Hey could induce the proliferation and apoptosis of VSMCs, which may be one of the mechanisms involved in atherogenesis by Hey.
