目的探讨水杨酸钠对豚鼠听反应阈值、螺旋神经节形态及γ氨基丁酸(GABA)、谷氨酸(GLU)表达的影响。方法将40只成年豚鼠随机均分为两组,A组予生理盐水腹腔注射,B组予水杨酸钠300 mg/(kg.d)腹腔注射;给药15 d后每组随机选取10只豚鼠,通过美国TDT系统测量两组听性脑干反应(ABR)阈值,扫描电镜观察两组豚鼠螺旋神经节形态学改变,采用免疫组织化学ABC法检测两组豚鼠耳蜗螺旋神经节GABA、GLU的表达。剩余动物停止给药,继续喂养30 d后,完成以上检测。结果 B组给药15 d后,豚鼠ABR阈值升高,螺旋神经节排列紊乱,胞体欠光滑,神经纤维易离断,螺旋神经节GABA、GLU的表达升高。B组停药30 d后,豚鼠ABR阈值仍停留在高阈值水平,螺旋神经节形态学上述改变仍存在,螺旋神经节GABA、GLU的表达较给药15 d后有所升高(P均〈0.05)。A组以上观察指标无明显变化。结论水杨酸钠可提高豚鼠听反应阈值,部分改变螺旋神经节形态,增高GABA、GLU的表达。
Objective To observe the effects of sodium salicylate on the auditory brainstem response and the mor- phology, the expressions of y-aminobutyricacid and glutamate of spiral ganglion neurons in guinea pigs. Methods Forty a- dult guinea pigs were randomly divided into two groups (20 cases for each) : the saline control group (group A ) and the salicylate group [ group B injected with a dose of 300 mg/( kg · d) for 15 days) ] respectively. After administration for 15 days, 10 guinea pigs were randomly chosen, then the rest of the animals were fed without adaministration for 30 days. Au- ditory brainstem response (detected by Tucker Davis Technologies), scanning electron microscopy, immunohistochemistry were used to evaluate the auditory function of guinea pigs, the morphological changes and the expressions of ~-aminobutyri- cacid and glutamate of the spiral ganglion neurons in guinea pigs. Results After administration for 15 days, ABR thresh- olds of guinea pigs increased, spiral ganglion was disorganized, cell body not smooth, nerve fiber apt to divide, the expres- sion of GABA, GLU of spiral ganglion increased in B group. After drug withdrawal for 30 days, ABR thresholds of guinea pigs was still at the high thresholds levels, spiral ganglion also exist aboved morphologically changes, the levels of GABA, GLU of spiral ganglion were higher than those after administration for 15 days (all P 〈 O. 05 ). But there was no obviously significance of the aboved indexes in group A. Conclusions Sodium salicylate could increase the the auditory brainstem response threshold of guinea pig and the expressions of ",/-aminobutyricacid and glutamate of the spiral ganglion neurons in guinea pigs. It could also change the morphologies of the spiral ganglion neurons partly.
