目的:探讨干扰素α-2b(IFN α-2b)对HepG 2.2.15细胞增殖、凋亡及HBsAg与HBeAg表达的影响。方法:应用SRB法检测IFNα-2b作用3、6d后对HepG 2.2.15细胞增殖的影响,ELISA法检测IFN α-2b对HBsAg与HBeAg表达的影响。Hochest 33342、PI双染观察细胞凋亡情况。结果:IFN α-2b作用3、6d后细胞增殖均有所抑制,3d后上清中HBsAg、HBeAg表达就受到抑制,并呈剂量依赖性,6d后HBsAg表达明显受抑,而HBeAg表达与3d后的结果相似。Hochest33342、PI双染显示IFNα-2b可引起细胞凋亡,随药物浓度增加,凋亡细胞也逐渐增加,并且有少量细胞开始死亡。结论:IFNα-2b体外可抑制HBsAg、HBeAg表达,且对细胞增殖也有所抑制,并可引起细胞的凋亡和死亡。
Objective To investigate the influences of interferon α-2b(IFN α-2b) on the proliferation,apoptosis,expression of HBsAg and HBeAg in HepG 2.2.15 cells.Methods After treated with IFN α-2b for 3 days or 6 days,the proliferation and apoptosis of HepG 2.2.15 cells were analyzed by SRB assay and double staining with Hochest 33342 and PI respectively.And the expression of HBsAg and HBeAg in treated HepG 2.2.15 cells were assayed by ELISA.Results IFN α-2b treatment inhibits the proliferation of HepG 2.2.15 cells and causes a G0/G1 cell cycle arrest.The expression of both HBsAg and HBeAg were inhibited in a dose-dependent manner after treated with IFN α-2b for 3 days.And the expression of HBsAg was further downregulated after 6 days,while the expression of HBeAg was similar to that of treated for 3 days.Hochest 33342 and PI double staining revealed that IFN α-2b also induced apoptosis in a concentration dependent manner.Conclusion IFN α-2b treatment inhibited not only the expression of HBsAg and HBeAg,but also the proliferation of HepG 2.2.15 cells and induced apoptosis in HepG 2.2.15 cells.
