目的检测新疆维、汉人群变应性鼻炎(AR)患者外周血CD4+淋巴细胞中IL-2及IL0mRNA的表达水平,比较2种基因在该疾病中的作用。方法应用实时荧光定量RT-PCR技术检测80例变应性鼻炎患者和80例正常对照者IL-2及IL-4mRNA的表达水平,其中鼻炎患者和对照者中维族、汉族各40例。结果选择管家基因GAPDH进行校正,IL-2mRNA表达水平在AR组为(8.66±1.92)coples/μl,维族和汉族分别为(8.52±2.15)和(8.81±1.79)coples/μl,AR对照组为(11.28±1.78)coples/μl,其中维族和汉族分别为(10.31±1.92)和(12.27±0.93)coples/μl.IL-4mRNA的表达水平经管家基因GAPDH校正后在AR组为(10.90±2.54)coples/μl,其中维族和汉族分别为(10.42±2.95)和(11.41±2.13)coples/μl,AR对照组为(3.07±1.18)coples/μl维族和汉族分别为(2.81±1.46)和(3.46±0.61)coples/μl。AR组IL-2mRNA的基因表达水平与健康对照者之间有差异(P〈0.05),维吾尔族和汉族之间无明显差异(p〉0.05)。AR组IL-4mRNA的基因表达水平高于健康对照者(P〈0.05),在维族汉族之间差异不大(P〉0.051。结论IL-2mRNA基因表达水平在AR组和健康对照者之间有显著差异(P〈0.05),IL-4mRNA的基因表达水平高于健康对照者。IL-2和IL-4基因在变应性鼻炎的发病过程中作用显著,IL-2和IL-4基因在维吾尔族和汉族AR患者之间无明显差异(P〉0.05)。
This study designed to investigate the expression levels of IL-2 and IL-4 mRNA in peripheral blood CD4+T lymphocyte of allergic rhinitis (AR) patients from Uighur and Han populations in Xinjiang, so as to compare the roles of the two genes in AR. Real-time quantitative RT-PCR was applied to detect the expression levels of IL-2 and IL-4 mRNA in 80 AR patients and 80 controls, including 40 Uighur and 40 Han in each group, with housekeeping genes GAPDH as correction gene. Result showed that mean level of IL-2 mRNA in AR group was 8.66±1.92, in which the Uighur and Han populations were 8.52±2.15 and 8.81±1.79 respectively. While the mean expression level of IL-2 mRNA in the control group was 11.28±1.78, in which Uighur and Han populations were 10.31±1.92 and 12.27±0.93, respectively. The mean expression level of IL-4 mRNA in AR group was 10.90±2.54, in which Uighur and Han populations were 10.42±2.95 and 11.41±2.13, respectively. While the mean expression level of IL4 mRNA in control group was 3.07±1.18, in which Uighur and Han populations were 2.81±1.46 and 3.46±0.61, respectively. The expression level of IL-2 mRNA was difference in AR patients and healthy controls (P 〈 0.05), but no significant difference in Uighur and Han AR patients (P〉0.05). The expression level of IL-4 mRNA in Han AR group was higher than that of the healthy controls (P〈 0.05), but no siginificant difference found between Uighur and Han AR patients (P〉 0.05). While the expression level of IL-2 mRNA showed significant difference between AR patients and healthy controls(P〈 0.05). Furthermore, the expression level of IL-4 mRNA in AR group was higher than that of healthy controls. In conclusion, IL-2 and IL-4 genes may play important roles in AR development. In addition IL-2 and 1L-4 genes level show no obvious difference in Uighur and Han AR patients(P〉0.05).
