目的:探讨益气消癥法中药对雌激素诱导子宫肌瘤模型豚鼠子宫组织增殖细胞核抗原(PCNA)、微血管密度(MVD)、碱性成纤维细胞生长因子(bFGF)蛋白表达的影响。方法:将去势豚鼠随机分为模型组、中药高、中、低剂量组及西药组,并以正常组作对照。通过去势加皮下注射苯甲酸雌二醇注射液建立子宫肌瘤模型,通过酶联免疫法检测各实验组子宫组织PCNA蛋白表达;免疫组化法检测MVD、bFGF蛋白表达。结果:模型组豚鼠子宫肌组织PCNA、MVD、bFGF蛋白表达显著高于正常对照组(P〈0.01,P〈0.05);中药各剂量组均有不同程度的降低,其中中药高剂量组豚鼠子宫组织PCNA、MVD、bFGF蛋白表达显著低于模型组(P〈0.01,P〈0.05),与西药组相比有显著性差异(P〈0.01,P〈0.05)。结论:益气消癥法中药可通过降调模型豚鼠子宫组织PCNA、MVD、bFGF蛋白表达,抑制血管生成和肌瘤细胞的增生,从而达治疗子宫肌瘤的作用。
Objective: To study the influence of Yiqi Xiaozheng method on the expression of MVD, PCNA, bFGF protein in estrogen-induced uterine fibroids tissue of guinea pig model. Methods: Ovariectomized guinea pigs were randomly divided into model group, high-dose group, medium-dose group, low-dose group and western medicine group (mifepristone), and the normal group was used as control. The castration and estrogen-induced methods were used to establish the uterine fibroids model. The expression of PCNA protein in each group was detected by enzyme-linked immunosorbent assay, and the protein expression of MVD and bFGF was detected by immunohistochemistry staining. Results: The expression of PCNA,MVD,bFGF protein in the model group were significantly higher than those in the normal group (P〈0.01). After treatment, the expression of PCNA, MVD, bFGF in Chinese medicine groups were decreased in different degrees. The expression of PCNA, MVD, bFGF in high-dose group was significantly lower than these in the model group (P〈0.01, P〈0.05), and compared with western medicine group, there also had significant difference (P〈0.01, P〈0.05). Conclusion: Yiqi Xiaozheng method could treat uterine fibroids through decreasing the expression of PCNA, MVD, bFGF protein in estrogen-induced uterine fibroids tissue of guinea pig model, and inhibiting angiogenesis and hyperplasia of leiomyoma cells.
