确定一种玉米须总DNA的提取方法.[方法]分别利用CTAB法和SDS法提取干燥和新鲜玉米须的总DNA,经过琼脂糖凝胶电泳进行定性检测,通过紫外分光光度法进行含量和纯度测定,确定适合玉米须DNA提取的方法.[结果]利用CTAB法和SDS法提取干燥玉米须的总DNA,产物呈淡黄色,经过琼脂糖凝胶电泳,几乎看不到明显的条带;利用上述两种方法提取新鲜玉米须的总DNA,电泳条带明显,CTAB法提取DNA浓度为11.5μg/m L,A_(260)/A_(280)为2.2,而SDS法提取DNA浓度为15.8μg/m L,A_(260)/A_(280)为1.9.[结论]新鲜玉米须中总DNA更容易提取;SDS法提取出的DNA浓度与纯度均优于CTAB法,更适宜用于新鲜玉米须DNA的提取.
[Objective]This study is aimed at screening a method to extract corn silk DNA.[Method] The CTAB method and SDS method were used to extract the DNA from dry and fresh corn silk. To determine the method,the DNA samples were detected by agarose gel electrophoresis and quantified by UV spectrophotometry.[Results] The DNA of dry and fresh corn silk was extracted by the above method,while the DNA sample from dry corn silk was slight yellow and showed no obvious bands of the yellow extraction in agarose gel electrophoresis; the fresh corn silk DNA present the clear band in gel.The DNA concentration was11.5 μg/m L and A_(260)/A_(280) was 2.2 by CTAB method,and the value of DNA by SDS method was 15.8 μg/m L and 1.9.[Conclusion] The fresh corn silk was easy to extract DNA; to the CTAB method,SDS method can provide the more high-quality DNA,so it was more suitable for DNA extraction from fresh corn silk.
