目的通过观察软肝升白颗粒对肝纤维化小鼠肝脏肝细胞生长因子及其C-Met mRNA表达的影响,探讨其抗肝纤维化的作用机制。方法 80只BABL/c小鼠给予0.03%硫代乙酰胺饮用水喂养20周,制备肝纤维化动物模型[1]。随机分为4组,每组20只。A组小鼠给予软肝升白颗粒11.2 g/(kg.d),B组小鼠给予软肝升白颗粒5.6 g/(kg.d),C组小鼠给予鳖甲软肝片1.12 g/(kg.d),D组分别给予等体积的生理盐水,每天灌胃1次,另外,20只小鼠作为正常对照组(E组),处理方法同D组。分别在4周和8周时,各处死小鼠10只,检测血清肝功能指标,各组取肝组织HE染色肝组织学观察,同时应用RT-PCR技术检测肝组织中肝细胞生长因子(HGF)及其受体C-Met mRNA的表达量。结果治疗前后肝功能和组织学均存在显著性差异。治疗组HGF/C-Met mRNA表达显著高于对照组(P〈0.05)。结论肝纤维化发生后,软肝升白颗粒能使HGF/C-Met mRNA高表达。
Objective To investigate the mechanism of Ruanganshengbai Granules in anti-hepatofibrosis through observing its influences on the expressions of hepatocyte growth factors(HGF) and C-Met-mRNA in mice with hepatic fibrosis.Methods BABL/c mice(n=80) were fed with 0.03% thioacetamide water for 20 weeks for establishing the model of hepatic fibrosis,and then all mice were randomly divided into four groups(each n=20).Group A was given Ruanganshengbai Granules [11.2 g/(kg·d)],group B was given Ruanganshengbai Granules [5.6 g/(kg·d)] and group C was given Biejiaruangan Tablets [1.12 g/(kg·d)].The model group(group D) and the control group(group E,n=20) were given equivalent normal saline solution once a day,respectively.On the 4th and 8th week,10 mice were killed in each group for detecting liver function indexes.The hepatic tissue were collected from all groups for histological observation after HE staining.The expressions of HGF and C-Met-mRNA were detected by using RT-PCR.Results There was significant difference in liver function indexes and histological observation before and after treatment.The expressions of HGF and C-Met-mRNA were higher significantly in the treatment groups than those in the control group(P0.05).Conclusion Ruanganshengbai Granules can increase the expressions of HGF and c-Met-mRNA after hepatic fibrosis occurred.
