目的:研究定心方(DXR)及氧化苦参碱(OMT)对大鼠缺血再灌注心律失常的保护作用及对细胞间黏附分子1(ICAM-1)表达的影响。方法:用冠脉结扎及再松开的方法制备缺血再灌注心律失常动物模型;记录Ⅱ导联心电图,对心律失常进行评分;双抗体夹心ABC-ELISA法检测血清中ICAM-1的表达。结果:定心方及氧化苦参碱干预后,缺血再灌注心律失常明显减轻(P〈0.01),血清中ICAM-1表达明显降低(P〈0.01)。结论:定心方及氧化苦参碱降低再灌注大鼠血清ICAM-1的表达,可能是其抗心律失常的机制之一。
Objective: To observe the effect of DingXin Recipe(DXR) and oxymatrine (OMT) on arrhythmia rats with myocardia ischemia - reperfusion injury and expression of ICAM - 1. Methods : Left - anterior coronary artery of rats was ligated for 15 minutes and then loosed for 30 minutes to establish the animal model of reperfusion arrhythmias. During the process, electrocardiogram was traced continuously to observe arrhythmias; Expression of serum ICAM - 1 was observed by means of Enzyme Linked- Immuno- Sorbent Assay. Results: DXR and OMT obviously attenuated reperfusion arrhythmias( P 〈 0.01 ), and relieved expression of serum ICAM - 1 ( P 〈 0.01 ). Conclusion : DXR and OMT can inhibit the activation of leucocyte and delivery of free radicals by attenuating expression of serum ICAM - 1, which can be one of the mechanisms of relieving reperfusion arrhythmias.
