以β-谷甾醇质量分数为评价指标,筛选白花丹野生变家种最佳栽培生长条件.采用HPLC-ELSD法测定在不同土壤、不同肥料、不同光照和不同植被下白花丹根、茎、叶中β-谷甾醇的质量分数.色谱条件:色谱柱为迪马C_(18)柱(250mm×4.6mm,5μm),流动相为纯甲醇,流速为1.0mL/min,柱温25℃;以蒸发光散射检测器进行检测,检测参数为漂移管温度65℃,载气(N_2)流速为2.5L/min.结果显示,β-谷甾醇在0.01~2.6μg范围内与峰面积值呈现良好的线性关系,回归方程为Y=1.500 5 X+5.683 6;β-谷甾醇在叶中质量分数最高,根中最低;在熟土和粘土中生长的药材质量分数高于沙土和生土;在30%光照条件下生长的药材质量分数最高.建立了白花丹中β-谷甾醇的HPLC-ELSD测定方法,采用此方法测定白花丹中β-谷甾醇的质量分数准确、可行.因此,以β-谷甾醇为主要活性成分的白花丹野生变家种最佳栽培条件为土壤选择熟土、沙土或粘土,光照条件选择30%或60%光照环境,或在林下栽培.
The content of β-sitosterol has been usedas evaluation index toscreen the best cultivation condi- tions of culitivatedHerba Plumbaginis Zeylanicae. In different soil, fertilizers, illumination and vegeta- tionhas been adopted. The higher performance liquid has been used to determine β-sitosterol content in roots, stems and leaves of home-grown HerbaPlumbaginis Zeylanicae. Diamondsil C18 (250 mm× 4.6 mm,5 μm) has also beenused with pure methanol as the mobile phase and the flow rate is 1.0 ml/min. The temperature of columnis 25℃. ELSD nebulization at 65℃ and flow rate of nitrogen at 2.5 L/min. The lin- ear ranges of β sitosterolare 0.1~2.6 μg/mL. The content of βsitosterol is highest in leaves and lowest in roots. The contents are higher in mellow and clay than that of sand and raw soil. Propagation of cuttingsis higher than other of reproduction material. The β-sitosterolis higher in herbs grown under 30% light con ditions. The determination method of β-sitosterol in Herba Plumbaginis Zeylanicaehas been establishedby HPLC-ELSD, the method of HPLC-ELSD is accurate and reliabl ba Plumbaginis Zeylanicae,mellow soil or clay soil, low light and e;considering β-sitosterol content of Her- hardwood forest were the best cultivatedconditions.
