目的:研究牵张应变诱导人牙周膜细胞(HPDLCs)凋亡过程中细胞凋亡相关基因Bcl-2和Bax的表达变化情况。方法:体外培养HPDLCs,利用动态机械应变细胞加载装置对HPDLCs加载20%动态牵张应变24h,通过real-time PCR检测Bcl-2和Bax在mRNA水平上的表达差异。结果:HPDLCs加载后,呈现栅栏状相互平行排列趋势,排列方向垂直于牵张力方向。Bcl-2和Bax基因表达显著增加,Bcl-2/Bax比值显著下降,有明显统计学意义(P〈0.05)。结论:牵张应变诱导人牙周膜细胞凋亡过程中,Bcl-2家族起到了重要作用。
Objective: The purpose of this study is to investigate the role of apoptosis related gene Bcl-2 and Bax in stretching force-induce HPDLCs. Methods: HPDLCs were cultured in vitro, and the cells were stretched by dynamic mechanical strain 20% for 24h. Real-time PCR was used to confirm the change of gene expression of Bcl-2 and Bax following stretching force. Results: HPDLCs inclined parallel to each other and aligned their long axis perpendicular to the stretching force vector. Bcl-2 and Bax significantly increased and the Bcl-2/Bax ratio showed a significant decline after application ofstretching force. Conclusion: The results demonstrated that Bcl-2 family plays an important role in stretching force-inducing HPDLCs apoptosis.
