1株来源于海绵的真菌(Aspergillus repens)其发酵产物的乙酸乙酯提取物表现出明显的细胞毒活性。本文采用溶剂萃取、硅胶柱层析及制备HPLC等分离手段,对该菌株发酵产物的活性部位进行了追踪分离,共分离得到3个生物碱成分。通过理化性质及利用波谱学手段,鉴定其结构分别为meleagrin(1)、neoechinulin A(2)和preechinulin(3)。采用SRB法初步评价了化合物1-3的抗肿瘤活性,结果表明,化合物1对P388和K562细胞表现出明显的细胞增殖抑制活性。
To explore the antitumor active component in fermentation of a sponge-derived fungus,Aspergillus repens,three compounds were isolated by using a bioassay guided fractionation against K562 cell line.Their structures were determined as meleagrin(1),neoechinulin A(2) and preechinulin(3) respectively on the bases of physico-chemical properties and spectral data.The proliferation inhibitoral activities of these compounds were evaluated by the SRB method against P388 and K562 cell lines.Compound 1 showed moderate inhibitory activity against both cell lines.
