激光显微切割技术已经成为一种能够快速稳定地从复杂组织获取均一样本的工具,该技术已经有了较为广泛的应用,并随着研究的不段深入,成为单细胞的分离和表达研究必不可少的应用工具。目前,使用激光显微切割分离细胞等研究已非常广泛,但在单条染色体分离的研究上,报道甚少,尤其在山羊上还没有激光显微切割的相关报道。因此,针对山羊单条染色体的分离进行细胞培养、膜玻片核型制备和激光显微切割等条件摸索,确定在ConA刺激剂作用下,培养48h后加入秋水仙素作用淋巴细胞5h,细胞生长良好,可以分离和获得较为理想的单条染色体。为大型哺乳动物的单条染色体分离研究提供借鉴。
Laser microdissection has become a fast and stable tool that can get the same type samples from the complex tissues.This technology has been more widely used in humans,plants and animals.With the development of the technology and deepening of research,it has become an essential tool for single cell separation and expression analysis.Currently it has been extensively used on microdissection of isolated cell research,but it was seldom reported on a single chromosome separation study,especially for laser microdissection of separated single chromosome in the goat.Therefore,this paper studied on the single chromosome separation from the cell culture,karyotype preparation on film slides and laser microdissection.The results show that cells grow well under the condition of stimulator ConA,culture 48 hours then adding the colchicine for 5hours;single chromosome can be obtained by the laser microdissection method.It will provide a useful reference for the study of single chromosome separation in large mammals.
