分离自废水中的食酸菌属菌株Q1能够在液固界面形成成熟的生物膜。利用转座子插入得到突变株△twbS,该突变株在生物膜形成功能缺失。经过序列分析,转座子插入基因与Acidovorax avenae sub sp.citrulli AAC00-1和Diaphorobacter sp.TPSY的感应激酶调控因子TwbS分别有60%和61%氨基酸同源性。进一步的研究表明该twbS基因与细菌的蹭行运动和Type IV菌毛的装配有关,从而影响细菌的初步粘附功能,最终达到调控生物膜形成。突变株△twbS可以通过回复突变实验恢复上述功能。
The waste water isolated tem of Acidovorax Q1 forms a substantial biofilm at the Liquid-Solid interface.One mutant,△twbS,leads to the identification of a biofilm-deficient derivative in which a transposon insertion disrupted a gene throush.BLASTP analysis,TwbS shares 60% and 61% amino acid identity with multi-sensor hybrid histidine kinase from Acidovorax avenae subsp.citrulli AAC00-1 and Diaphorobacter sp.TPSY,respectively.Further examination showed that the twbS mutant was relgvant to twitching motility defective and Type IV pili assembly defective.Thus twbs influenled the preliminary adherent function,the formation of biofilin can be regnlated.The biofilm formation defect in the twbS mutant was complemented by providing the twbS gene.
