目的观察苓桂术甘汤对慢性心衰竭(chronic heart failure,CHF)大鼠血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)、内皮素-1(endothelin-1,ET-1)及肿瘤坏死因子-α(tumor necrosis factor-α,TNF—α)、白细胞介素-1β((interleukin-1,IL-1β)的影响。方法采用冠状动脉结扎法复制CHF大鼠模型,于模型复制后第7周将大鼠随机分成模型组,卡托普利组,苓桂术甘汤小(4.29g/kg)、中(21.45g/kg)、大(42.90g/kg)剂量组,另设假术组,分别灌胃给药,连续给药4周后,腹主动脉采血,采用放射免疫法测定各组大鼠血清AngⅡ、ET-1、TNF—α和IL-1β水平。结果苓桂术甘汤小、中、大剂量组与卡托普利组AngⅡ、ET-1、TNF—α和IL-1β水平显著下降,与模型组比较,差异具有显著性(P〈0.01);苓桂术甘汤大剂量组TNF—α、IL-1β水平显著低于卡托普利组(P〈0.05)。结论苓桂术甘汤能明显降低CHF大鼠血清AngⅡ、ET-1、TNF—α和IL-1β水平,该方阻抑CHF大鼠心室重构,改善CHF大鼠心脏舒缩性能作用与其抑制神经内分泌及细胞因子过度表达密切相关。
Objective To observe effects of Linggui Zhugan Decoction, a traditional Chinese prescription, on levels of angiotensin Ⅱ (Ang Ⅱ ), endothelin-1 (ET-1), tumor necrosis factor-α (TNF -α) and interleukin 1β (IL-1β) in rats with chronic heart failure (CHF). Methods The CHF model was induced by ligation of the anterior descending coronary artery. From the 7th week after the operation, the rats with CHF were intragastrically administered with low (4. 29 g/kg), middle (21. 45 g/kg) and high (42. 90 g/kg) dosage of Linggu Zhugan Decoction or captopril, continuously during 4 weeks, and then serum levels of Ang Ⅱ, ET-1, TNF-α and IL-1β of rats in all groups were detected by radioimmunoassay. Results Compared with those in the model group, serum contents of Ang Ⅱ , ET-1, TNF-a and IL-1β in groups treated with low, middle and high dosage of Linggui Zhugan Decoction and captopril were significantly decreased (P〈0.01). Serum levels of TNF-α and IL-1β in the group treated with high dosage of Linggui Zhugan Decoction were significantly lower than those in captopril group (P〈0.05). Conclusion Linggui Zhugan Decoction can obviously decrease serum levels of Ang Ⅱ, ET-1, TNF-α and IL-1β in rats with CHF, its inhibitive effect on ventricle reconstitution and its improving the systolic and diastolic function of CHF rat heart might be related with inhibiting the overactivity of neuroendocrine'and cytokines.
