蚕丝纤维作为一种天然材料,是最早被利用的生物材料之一,其中含有的丝素蛋白使其具有优良的力学性能,组织相容性及可降解性,在生物医学领域得到广泛关注.将丝素蛋白从蚕丝中提取出来,制成纳米球,使其负载内皮细胞生长因子(VEGF)并混入聚己内酯(PCL)纺丝液中,通过静电纺丝技术制备得到纤维内含有丝素纳米球的PCL电纺膜.体外释放表明丝素纳米球及含丝素纳米球的电纺纤维膜都有很好的缓释效果;生物体内评价结果表明负载有VEGF的丝素纳米球混合的PCL电纺支架促进了体内的细胞浸润和周围血管化.
Silk fiber is a kind of natural materials,and it is one of the earliest biological materials.Silk fiborin is the main component of the silk fiber,and it contribute to the mechanical property,tissue compatibility and degradable property.In this paper,silk fiborin was extracted from the silk fiber and made into nanoparticles,then loaded with vascular endothelial growth factor and mixed into the PCL electrostatic spinning solution.The nanofiber film of PCL combined with silk fiborin can be obtained ith electrospinnmg technology.The in vitro release of VEGF indicate that the silk fiborin nanoparticles and films with silk fiborin nanoparticles show sustained-release effect;the evaluation of in vivo study indicate that the PCL scaffold combined with VEGF-silk fiborin nanoparticles promote the cell infiltration and vascularization.
