目的观察激动乙醛脱氢酶2(ALDH2)是否能对抗糖尿病大鼠诱导的肺损伤,并分析相关机制。方法雄性sD大鼠分为正常对照组、糖尿病组和糖尿病+ALDH2激动剂乙醇组。8w后测定大鼠空腹血糖水平和肺系数(肺湿重(g)/体重(g)×100],分光光度法测定肺组织SOD活性和MDA含量。Western印迹测定肺组织ALDH2蛋白表达。结果与对照组相比,糖尿病组大鼠空腹血糖明显升高,体重降低,肺系数增加,肺组织SOD活性下降,MDA含量增加,ALDH2蛋白表达降低(均P〈0.01)。与糖尿病组大鼠相比,糖尿病+乙醇组大鼠空腹血糖降低,体重增加,肺系数降低,肺组织SOD活性升高,MDA含量下降,ALDH2蛋白表达增加(均P〈0.05或P〈0.01)。结论糖尿病大鼠诱导的肺损伤中,肺组织AL-DI-12可表达降低,可能与氧化应激损伤加重有关。激动ALDH2可减弱糖尿病诱导的肺损伤。
Objeetlve To observe whether activation of ALDH2 can alleviate lung injury induced by diabetic, and analyze the likely mechanism. Methods Male SD rats were divided into control, diabetic rat (DM) and EtOH + DM groups. The levels of fasting blood glu- cose (FPG) and the ratio of lung weight to body weight (LW/BW) were measured. The MDA content and SOD activity in lung tissue were measured. Pulmonary ALDH2 protein expression was detected by Western blot. Results Compared with that of control group, the level of FPG was increased, body weight was decreased while LW/BW was increased in DM group. Lung MDA content was increased, and SOD ac- tivity was decreased. ALDH2 protein expression was decreased. Compared with those of DM group, the levels of FPG, LW/BW were decreased, body weight was increased, MDA content was decreased, SOD activity and ALDH2 protein expression were increased in EtOH + DM group. Conclusions In diabetic rat, pulmonary ALDH2 expression is decreased accompanying with the happening of lung oxidative stress. Activation of ALDH2 could decrease lung injury induced by diabetes.
