探讨超声结合原卟啉Ⅸ(PPⅨ)对S180肿瘤细胞膜功能的影响及其作用机制。采用频率为1.1 MHz,强度为3 W/cm2的聚焦超声,结合1μg/mL的PPⅨ作用于S180肿瘤细胞,借助于流式细胞仪,采用荧光探针DCFH-DA(2',7'-二氯双氢荧光素双乙酸酯)、FD500(异硫氰酸荧光素钠标记的右旋糖酐)、DiBAC4(3)和Fluo3-AM分别检测胞内活性氧生成量、细胞膜通透性、质膜电位及胞内游离Ca2+的浓度变化。研究结果表明,超声结合PPⅨ比单纯超声或单纯PPⅨ对细胞膜功能损伤作用都强,这种损伤作用可能与影响质膜功能的活性氧含量、细胞膜通透性、质膜电位以及胞内游离Ca2+浓度变化相关。
This study is purposed to investigate the effect of cell membrane function induced by ultrasound activating ProtoporphyrinⅨ(PPⅨ) on S180 tumor cells.Ultrasound at the frequency of 1.1 MHz and intensity of 3 W /cm2,combined with 1 μg /mL PP Ⅸ,acted on S180 tumor cells.The production of intracellular reactive oxygen species,cell membrane permeability,plasma membrane potential,and intracellular Ca2 + concentration were assessed using fluorescent dyes DCFH-DA(2′,7′-dichlorodihydrofluorescein-diacetate),FD500(fluorescein isothiocyanate-dextran),DiBAC4(3) and Fluo3-AM respectively under a flow cytometry.The results showed that ultrasound activating PPⅨ strongly impaired the membrane function of S180 cells,than the ultrasound or the PPⅨ alone.The damage might be induced by the generation of reactive oxygen species,permeability of cell membrane,plasma membrane potential and intracellular Ca2 + concentration,which have some relation with the membrane function.
