目的:探讨长链非编码RNAH19和HOTAIR在宫颈癌血清中的表达及其临床意义。方法:收集52例宫颈癌术前血清标本,同时选取年龄配对的39例因妇科良性疾病入院宫颈癌筛查正常的患者血清标本作为对照组。其中10例宫颈癌患者行术前、术后对比。用实时定量RT-PCR(qRT-PCR)检测上述标本的LncRNA H19和HOTAIR表达水平,并对宫颈癌患者术前、术后进行对比。分析宫颈癌患者血清LncRNA H19和HOTAIR与其临床病理参数的关系,采用受试者工作曲线(ROC)对血清LncRNA H19和HOTAIR诊断效能进行评价。结果:宫颈癌患者血清LncRNA H19和HOTAIR表达水平较对照组显著升高,且术后较术前表达显著下降,差异有统计学意义(P〈0.05);宫颈癌患者血清中H19和HOTAIR的表达水平与患者年龄、病理类型、临床分期、SCC均无相关性(P〈0.05);血清LncRNA H19单独诊断宫颈癌的ROC曲线下面积(AUC)、灵敏度和特异度分别为0.661、30.8%和94.9%;血清LncRNA HOTAIR单独诊断宫颈癌的AUC、灵敏度和特异度分别为0.707、69.2%和66.7%。结论:血清LncRNA H19和HOTAIR可以作为宫颈癌临床诊断和术后监测潜在的循环标记物。
Objective:To investigate the expression and clinical significance of long non-coding RNA H19 and HOTAIR in cervical cancer.Methods:Preoperative serum samples were collected from 52 patients with cervical cancer.At the same time,the serum samples of 39 patients with gynecological benign diseases but normal cervical cancer screening were selected as control group.Among them,10 patients with cervical cancer underwent preoperative and postoperative comparison.LncRNA H19 and HOTAIR expression were detected by Real time quantitative RT-PCR (qRT-PCR) in all samples and among them patients with cervical cancer were compared before and after operation.The relationship between H19 and HOTAIR expression in cervical cancer patients serum and clinical pathological parameters were further analysed and diagnostic efficacy of serum LncRNA H19 and HOTAIR were evaluated by the receiver operating characteristic curve (ROC).Results:H19 and HOTAIR expression in serum of patients with cervical cancer were significantly higher than those in the control group and decreased significantly after operation (P〈0.05).And they were not correlated with age,pathological type,clinical stage and SCC (P〈0.05).The area under the ROC curve (AUC) and the sensitivity and specificity of serum LncRNA H19 in the diagnosis of cervical cancer were 0.661,30.8% and 94.9% respectively and were 0.707,69.2% and 66.7% for HOTAIR respectively.Conclusion:Serum LncRNA H19 and HOTAIR may be a potential marker for clinical diagnosis and postoperative monitoring of cervical cancer.
