目的:探讨ETK调节膀胱癌细胞侵袭转移的分子机制。方法:首先应用Westernblot技术检测ETK在5种膀胱癌细胞中的表达水平,再使用ETKshRNA技术下调膀胱癌细胞ETK蛋白表达,然后应用侵袭试验检测ETK对膀胱癌细胞侵袭转移能力的影响,最后以Westernblot检测p-AKT,AKT,p-STAT-3和STAT-3的表达水平。结果:5种膀胱癌细胞均表达ETK蛋白,其中以T24和UM-UC-3表达水平最高,下调ETK表达可以抑制膀胱癌细胞T24和UM-UC-3的迁移和侵袭能力,并与AKT和STAT-3的磷酸化水平下降有关。结论:ETK在膀胱癌的迁移和侵袭作用中具有重要的作用,因而ETK可能具有成为抑制膀胱癌进展潜在的新的治疗靶点的价值。
Objective To investigate the molecular mechanism of ETK regulating the metastasis and invasion of bladder transitional cell carcinoma(TCC).Methods Expression of ETK in 5 kinds of bladder cancer cell lines was validated by Western blot.The expression of ETK protein in bladder cancer cells was down-regulated by ETK shRNA method.The effects of ETK on the metastasis and invasion of bladder cancer cells were assessed by Transwell assay,and finally the expressions of p-AKT,AKT,p-STAT-3 and SATA-3 were tested by Western blot.Results ETK was expressed in all 5 kinds of bladder cancer cell lines,especially in T24 and UM-UC-3.Moreover,the down-regulation of ETK could significantly inhibit the metastasis and invasion of T24 and UM-UC-3 cells,and was related to the reduction of p-AKT and p-STAT-3.Conclusions ETK may play a role in the metastasis and invasion of bladder cancer,which may be a potential target for novel therapy for bladder cancer.
