以15个彩色马蹄莲栽培品种的无菌苗为材料,接种至筛选出的最适丛生芽诱导培养基MS+0.1 mg/LNAA+1.5mg/L6-BA中,分别比较了它们丛生芽诱导分化能力和秋水仙素诱导无性多倍体产生的差异以及各品种多倍体与二倍体早期形态学的差异.结果表明:彩色马蹄莲无性多倍体的成功诱导与点状丛生芽块的形成密切相关,‘ Treasure’,‘Pot of gold’,‘Liberty’,‘Black Magic’,‘Gold Finger'5个品种成功获得多倍体,丛生芽的增殖系数分别为3.7,4.7,5.6,4.3,5.2,秋水仙素处理后多倍体的变异率分别表现为16%,22%,28%,20%,25%;5个品种多倍体幼苗的叶长分别为二倍体的123.9%,119.8%,107.9%,108.8,142.9%,叶宽分别为二倍体的207.5%,189.5%,171.7%,173.3%和157.1%.
The in vitro plantlets of 15 colored Zantedeschia hybrid cultivated varieties were inoculated in the media:MS + NAA 0.1 mg/L + 6-BA 1.5mg/L,and compared their difference on shoot buds induction and asexual tetraploids induction by colchicine treatment; the morphological differences between asexual tetraploids and diploids in the young plant stage was analyzed.The result showed that:five varieties,'Treasure',' Pot of gold',' Liberty',' Black Magic',' Gold Finger' were induced to produce the tetraploids successfully,and polyploid induction in colored Zantedeschia hybrid had a high relation to the formation of very young cluster buds like dots.The polyploid induction was positive to the multiplication ratio of cluster buds.The multiplication ratios of cluster buds were 3.7,4.7,5.6,4.3,5.2 and the polyploid variation rates by colchicine treatment were 16%,22%,28%,20%,25% respectively.Compared with diploid,the leaf lengths of polyploid variation of five varieties seedlings were 123.9%,119.8%,107.9%,108.8,142.9%,and the leaf width was 207.5%,189.5%,171.7%,173.3% and 157.1%,respectively.
