目的:对狼毒大戟中没食子酸乙酯进行提取分离,并探索其抗肿瘤作用。方法:采用硅胶柱色谱法、Sephadex LH-20凝胶柱色谱法和半制备高效液相色谱法从狼毒大戟的乙酸乙酯层分离纯化没食子酸乙酯,通过MTT法观察其在不同浓度和时间条件下对三种肿瘤细胞的生长抑制作用。结果:没食子酸乙酯对结肠癌LOVO细胞、肺癌A549细胞、口腔鳞癌CAL-27细胞的生长均具有一定的抑制作用,经药物作用72h后,最大抑制率分别达到(74.67±0.47)%、(62.01±1.27)%和(37.17±2.12)%。结论:狼毒大戟乙酸乙酯层提取分离的没食子酸乙酯在体外有抗肿瘤活性。
Objective: To extract and isolate ethyl gallate from Euphorbia fischeriana and to explore its antineoplastic activity. Method: Isolate ethyl gallate from the ethyl acetate parts of Euphorbia fischeriana by silica gel column chromatography,Sephadex LH-20 chromatography and semi-preparative reversed phase liquid chromatography. MTT assay was used to test the inhibitive effect of ethyl gallate on three tumor cells with different concertrations and at different action time. Results: The results showed that ethyl gallate inhibited the growth in colon cancer cells LOVO,lung cancer cells A549 and oral adenosquamous carcinoma cells CAL-27. After they were treated with different concentrations of ethyl gallate for 72h,maximum inhibitive rates were( 74. 67 ± 0. 47) %,( 62. 01 ± 1. 27) % and( 37. 17 ± 2. 12) % respectively. Conclusion: Ethyl gallate from Euphorbia fischeriana has an anti-tumor effect in vitro.
