从腾冲热海温泉中分离嗜热噬菌体,并初步分析其特征.采用双层平板法分离纯化嗜热噬菌体,对分离得到的噬菌体进行电镜形态观察和生理生化分析.结果从腾冲热海温泉中分离得到的噬菌体为长杆状;其感染宿主菌N J2形成清晰的噬菌斑,最适感染温度为55℃,最适感染pH值为7.5.该噬菌体对氯仿不敏感;但其对非离子去垢剂SDS和对蛋白酶K敏感.将这株噬菌体命名为嗜热芽孢杆菌杆状噬菌体TBRP3(Thermophilic bacillus rod-shaped phage 3).
The aim is to separate thermophilic bacteriophage in the spring from Tengchong hot sea, and analy zes its features. Doublelayer plate is used to separate and purify thermophilic bacteriophage, taken electron mi croscopy morphology on it, taking a physiological and biochemical analysis on the obtained thermophilic bacterio phage as well. The result is that the thermophilic bacteriophage obtained is of long rod, and its host N J2 forms clear plague. The most effective inflected temperature is 55 ℃, and the pH at 7.5. The bacteriophage is not sensitive to chloroform but to SDS and protease K. And the bacteriophage is named as TBRP3 ( Thermophilic bacillus rodshaped phaze ).
