In order to establish a new method for the PBBs detection by immuno-PCR, two sorts polybrominated biphenyls hapten was synthesized by introducing amino group and carboxyl group as a substituent at the aromatic ring while retaining the biphenyls ring. The hapten was conjugated to BSA via amino diazotization linkage and by the NHS ester method to get immunogen. The conjugation products was identified by ultraviolet spectrophotometry. The results showed that the molar incorporation ratio reached 1:17 and 1: 19. New Zeal- and rabbits were used to injected immunogens to validate the haptens. The antiserum exhibited relatively high antibody titres (1: 32-1: 64) in double agar diffusion.
