为研究老鹳草、大血藤、吴茱萸、锁阳及木瓜5种中药材提取物对黄嘌呤氧化酶的抑制作用.以别嘌呤醇作为阳性对照,采用紫外分光光度法在体外直接测定5种中药材不同浓度的提取物对黄嘌呤氧化酶的抑制作用.结果显示:5种中药材提取物(20 mg/mL)对黄嘌呤氧化酶的抑制率均大于50%,老鹳草的醇提物和水提物对黄嘌呤氧化酶的抑制作用最明显,其IC50分别为3.141 mg/mL、2.407 mg/mL.除老鹳草外,其余4种中药材的醇提物的IC50均小于水提物的IC50.表明5种中药材的醇提物和水提物对黄嘌呤氧化酶均具有较好的抑制作用,老鹳草的提取物抑制作用最明显.
The aim of the project is to study the inhibitory effects on xanthine oxidase by the extracts from five Chinese herbal medicines:Geranium wilfordii Maxim, Sargentodoxa cuneata (Oliv.) Rehd. et Wils, Evodia rutaecarpa (Juss.) Benth, Cynomorium songaricum Rupr and Chaenomeles sinensis (Thouin) Koehne. We directly measure the inhibitory effects on xanthine oxidase by the extracts of different concentrations from five Chinese herbal medicines using UV spectrophotometry in vitro, with allopurinol as positive control. The inhi-bition rate on the xanthine oxidase by the extracts (20 mg/mL) from all five Chinese herbal medicines is greater than 50%. The inhibition on xanthine oxidase by the ethanol extracts and water extracts of Geranium wilfordii is the most obvious and the IC50 are 3. 141 mg/mL and 2. 407 mg/mL, respectively. Apart from Ge-ranium wilfordii, the IC50 of the ethanol extracts of remaining four Chinese herbal medicines is less than the IC50 of the water extracts. It is concluded that all the ethanol extracts and water extracts of the five Chinese herbal medicines have good inhibition on xanthine oxidase, with the inhibition by Geranium wilfordii extracts the most obvious.
