目的:研究玻连蛋白(vitronectin,Vn)在肺血栓栓塞(pulmonary thromboembolism,PTE)中国大耳白兔肺动脉内皮中的表达变化,以及Vn在肺血栓栓塞病理过程中的作用。方法:将28只健康成年中国大耳白兔随机分为观察组和对照组,经股静脉分别注射自体血栓和生理盐水。各组于注射后1、3、5、7 d处死动物并取肺动脉及肺组织。分别进行苏木精-伊红(hematoxylin and eosin,H&E)染色、免疫组织化学染色及实时荧光定量聚合酶链式反应(real-time quantitative polymerase chain reaction,qRT-PCR)检测,观察肺组织及肺血管结构、定位及定量分析Vn在肺动脉中表达情况。结果:H&E染色显示观察组动脉内血栓栓塞,而对照组肺组织及肺动脉正常。免疫组织化学染色表明Vn表达于肺动脉内皮;qRT-PCR结果表明观察组肺动脉内皮中Vn表达量均高于对照组(P〈0.05),且在第3天达峰值(P〈0.05)。结论:Vn在肺血栓栓塞白兔肺动脉内皮中表达水平增高,Vn可能参与了肺血栓栓塞病程中的血栓形成过程。
Objective: To investigate the variation of Vitronectin(Vn) expression level in pulmonary artery endothelium of big-ear white rabbits with pulmonary thromboembolism(PTE) and the role of Vn in pulmonary thromboembolism. Methods:28 healthy adult big-ear white rabbits were randomly divided into the observation group and the control group. Their femoral veins were detached and injected intravenously with autologous blood clots or mere saline. Rabbits were killed on 1, 3, 5 and7 days after injection. Their lung tissues together with pulmonary arteries were harvested and processed for hematoxylin and eosin( HE) staining, immunohistochemistry and real-time quantitative polymerase chain reaction(q RT-PCR) analysis.Results: HE staining slides show normal structure of lungs in the control group, but thrombi in the arteries of the observation group. The distribution of Vn was confirmed in the endothelium of pulmonary artery by immunohistochemistry staining. q RTPCR results show Vn m RNA was over-expressed in pulmonary arteries in the observation group(P0.05). The expression level was highest on the third day after injection with clots(P0.05). Conclusions: Vn over-expressed in the endothelium of PTE rabbits. It may participate in the progress of pulmonary thromboembolism.
