目的研究不同植物生长调节剂组合和浓度配比对诱导福建金线莲丛生芽增殖的影响及金线莲组培苗芽发生及生长发育过程。方法以MS为基本培养基,附加9种植物生长调节剂组合,以金线莲无菌的茎段或茎尖为外植体,诱导腋芽增殖,定期观察记录芽发生过程,芽增殖情况,株高等生长状态指标。在筛选出适宜的植物生长调节剂组合基础上进一步考察9种浓度配比对诱导芽增殖的影响,并培育成苗。结果含BA+生长素培养基诱导茎尖、茎段芽增殖数高于含KT+生长素、ZT+生长素培养基。其中在BA+NAA培养基上茎尖、茎段芽增殖数最高,分别为对照的3.62和3.56倍。在BA+IBA的培养基上诱导茎段产生丛芽数最多(14丛)。在BA+生长素培养基上的苗比在基本培养基、KT+生长素、ZT+生长素培养基上的苗粗壮,节间缩短,根短粗。在BA+NAA组合中,以BA2.0mg·L^-1+NAA2.0mg·L^-1的浓度配比诱导茎段芽增殖效果较好,为对照的5.29倍。再生苗转接到培养基中10d后即可生根,25d成为完整苗。结论在特定浓度下,不同植物生长调节剂组合诱导芽增殖的效果是不同的,其中细胞分裂素对诱导芽增殖起主要作用,MS培养基中附加BA与NAA或IBA的组合对诱导金线莲芽增殖具有较好效果。而当BA与NAA组合及在本实验供试浓度范围内,NAA是芽增殖的主要影响因素。
OBJECTIVE To study the effect of different combinations and concentrations of plant growth regulators on proliferation of axillary buds of Anoectochilus roxburghii. And to observe the course of growth and development of buds and formation of complete plantlets. METHODS Shoot tips and nodal segments were used for axillary shoot proliferation on MS basal medium supplemented with nine kinds of combinations of cytokinin ( 1 mg · L^-1 ) and auxin (0. 5 mg· L^-1 ). Some growth index, such as the course of buds formation and development, buds proliferation, height of buds and shoots, were observed routinely. The effect of different concentration of plant growth regulators on axillary buds proliferation of A. roxburghii was carried out based on screening the suitable regulator combination. RESULTS Axillary buds induction on BA + auxin media was higher than that on MS, KT + auxin and ZT + auxin media. There are usally 2 -4 level lateral buds ramifications in media containing BA, higher than other media, The number of buds proliferation per nodal segment and shoot tip induced on BA + NAA medium were maximum, which were 3.62 times and 3.56 times as control, respectively. Multiple buds induction of nodal segments was the highest ( 14 clusters) on BA + IBA medium. Buds growing on BA + auxin media were robust, its intersegments were shorten and roots were short and thick. The combination of BA 2. 0 mg· L^-1 + NAA 2. 0 mg· L^-1 generated the good effect on axillary buds proliferation of A. roxburghii, which was 5.29 times as control. Buds (2 -2. 5 cm long) were cut and transfered into MS medium. Complete plantlets were formed within 25 d. CONCLUSION Different combinations of plant growth regulators with specific concentration have different effects on buds induction, while cytokinin has a major effect. Combinations of BA + NAA and BA + IBA are the optimum media for micropropagation of A. roxburghii. Compared with BA, NAA showed a major effect on the number of buds proliferation concentratio
