为了研究猪肌内脂肪(IMF)含量与相关基因多态性的关系,试验采用PCR—SSCP技术检测了75头大河乌猪和41头长白猪脂肪细胞型脂肪酸结合蛋白(A—FABP)基因的第1内含子区的遗传变异,并用最小二乘法模型分析了A—FABP基因对猪肌内脂肪含量的遗传效应。结果表明:在大河乌猪和长白猪A—FABP基因第1内含子中均存在多态性,共发现5个等位基因(A、B、C、D、E),6种基因型(AB基因型、BB基因型、BC基因型、BD基因型、CC基因型、CE基因型)。大河乌猪肌内脂肪含量BD基因型显著高于其他5种基因型(P〈0.05),BC基因型显著低于其他5种基因型(P〈0.05),BD基因型与其他5种基因型之间最小二乘均值差异显著(P〈0.05),BD基因型对PCR—SS-CP的最小二乘均值最大,证明BD基因型在该位点对肌内脂肪含量的效应值最大。长白猪肌内脂肪含量CC基因型高于其他5种基因型,BC基因型显著低于其他5种基因型(P〈0.05),最小二乘均值6种基因型之间差异均不显著(P〉0.05),CC基因型对PCR—SSCP的最小二乘均值最大,证明CC基因型在该位点对肌内脂肪含量的效应值最大。因此,可以通过提高BD基因型的频率来增加大河乌猪肌内脂肪含量,达到改善大河乌猪猪肉品质的目的。
To investigate the correlation betweenthe intramuscular fat (IMF) of pig and the pnlymorphism - related gene. Polymorphism of the intron 1 on A - FABP gene of 75 Dahe Black Pig and 41 Landrace were tested by using the PCR - single strand conformation polymorphism (PCR- SSCP), and then the relations of A - FABP gene to intramuscular fat contents (IMF) were analyzed by the general linear model (GLM). The results showed that the polymorphism existed in two kinds of pigs on A - FABP gene in the intron 1, and there were 5 alleles ( A, B, C, D, and E) ; 6 genotypes ( AB, BB, BC, BD, CC, and CE) , in which the genotype of BD of A - FABP was higher than the other five genotypes in Dahe Black Pig intramuscular fat content; the intramuscular fat content were significantly different' ( P 〈 O. 05 ). The genotype of BC was significantly lower than the other five genotypes in Dahe Black Pig intramuscular fat content (P 〈 O. 05 ) , and the least squares mean difference between BD and other five genotypes significantly ( P 〈0. 05 ) and the genotype of BD least squares maximum value PCR - SSCP. this data indicated that the genotype was most important factor for the locus effect value of intramuscular fat content. The genotype of CC was higher than the other five genotypes in Landrace intramuscular fat content, but intramuscular fat content was not significantly different; the geno- type of BC of A - FABP was significantly lower than the other five genotypes in Landraee intramuscular fat content ( P 〈 0.05 ) ; six genotypes were not significant in the least squares mean differences ( P〉0. 05 ) ; the genotype of CC least squares maximum value PCR - SSCP revealed that the genotype in this locus effect value of intramuscular fat content was also biggest. It is suggested that we can increase the genotype fre-quencies of BD to increase the intramuscular fat content of Dahe Black Pig for improving the quality of Dahe Black Pigs.
