在猪链球菌毒力相关基因orf2和mrp之间发现一个新的开放阅读框,经酶活性染色试验证实该阅读框编码的蛋白具有次黄嘌呤核苷酸脱氢酶(IMPDH)活性,该阅读框共计957 bp,编码319个氨基酸。经Interpro和PHD软件分析,推测该蛋白的功能结构域存在于9~798 bp基因片段所编码的多肽中,特对这790个碱基依次缺失,以找寻酶的催化部位和底物结合部位。4个缺失基因和全长基因经过基因克隆,表达出5种相应的蛋白。免疫印迹证实这5种蛋白可被猪链球菌2型(SS2)的抗血清识别,且其中的4种蛋白含有IMPDH单抗(1A8)识别表位;通过活性染色,初步确定了SS2中IMDPH的催化部位和底物结合部位。
