目的 研究抗CD81抗体作用星形胶质细胞不同时间后对细胞增殖的影响。方法 纯化的星形胶质细胞培养12h后,加入抗CD81抗体(4ug/ml),分别作用18、32及96h后,采用MTT比色法检测星形胶质细胞的细胞活性,计算细胞抑制率;用流式细胞仪检测细胞周期时相。结果 MTT法检测结果显示与未作用细胞相比,抗CD81抗体作用18、32、96h后,星形胶质细胞增殖显著受抑制,其抑制率分别为11.17%、13.17%、27.49%。流式细胞术检测结果显示抗CD81抗体作用18h后,S期细胞指数与未作用细胞相比,增加了16.09,星形胶质细胞主要被阻滞于S期;作用32h后,G0/G1期细胞指数与未作用细胞相比增加了4.08;96h后,G0/G1期细胞指数与未作用细胞相比增加了5.87,细胞主要滞留于G0/G1期。结论 抗CD81抗体作用不同时间均对星形胶质细胞有显著抑制作用,抑制作用随作用时间延长而增强。早期,抗CD81抗体使星形胶质细胞阻滞于S期,随着作用时间的延长,逐渐使细胞阻滞于G0/G1期。
Objective To investigate the inhibitory effects of anti-CD81 (antibodies against CD81)on astrocyte proliferation. Methods Purified astrocytes from newborn rats cerebral cortex were cultured for 12 h and incubated with anti-CD81 antibody (4 ug/ml) for 18, 32 and 96 h respectively, The viability of astrocytes was tested by MTT and the inhibition rate of astrocytes was calculated. The cell cycles of the astrocytes were measured by flow cytometery. Results MTT results revealed that after treatment with anti-CD81 for 18, 32 and 96 h respectively, the proliferation of astrocytes was significantly inhibited as compared with control groups with the inhibition rate of astrocytes being 11.17%, 13.17% and 27.49% respectively. Cell cycle analysis showed that the cells were arrested in S phase with the index of cells in S phase being increased by 16.09 after treatment with anti-CDS1 antibody for 18 h, and the cells were arrested in G0/G1 phase with the index of cells in G0/G1 phase being increased by 4.08 at 32 h and 5.87 at 96 h respectively. Conclusion Anti-CD81 antibody remarkably inhibits the proliferation of astrocytes in a time-dependent manner. The cells are arrested in S phase at 18 h and in G0/G1 phase as time goes on.
