目的探讨catenin家族的重要成员p120-catenin(p120ctn)对E—cadherin介导的细胞粘附作用的调节及机制,检测在活体内外肺癌细胞系中p120ctn对E—cadherin/β—catenin复合体的调节作用。及其对肺癌细胞侵袭和转移能力的影响。方法应用siRNA技术建立p120ctn基因沉默的多种癌细胞系后,采用Western Blot,Transwell分析,裸鼠移植实验等方法进行检测。结果在成功干扰p120ctn表达的肺癌细胞系中,应用Western Blot和RT—PCR方法,我们观察到E—cadherin的蛋白表达出现不同程度的减弱,但mRNA的表达几乎没有变化;而β-catenin在蛋白和mRNA水平上都出现了明显的表达减弱。同时应用Transwell和裸鼠移植实验,我们在活体内外还观察到p120ctn缺失的肺癌细胞系具有较强的侵袭转移能力。结论肺癌细胞系中p120ctn的表达缺失可能通过下调E—cadhefin和β-catenin的表达,参与肺癌细胞的侵袭和转移。
Objective To investigate the role and possible mechanism of p120-catenin ( p120ctn ) in lung cancer. Methods We knocked down p120ctn using siRNA, and test the influence on E-cadherin complex by Western Blot, Transwell assay, and Transplanted nude mice. Results Using Western Blot and RT-PCR, we found that pl20ctn siRNA reduced the expression of E-cadherin at protein level and β-catenin both at protein and mRNA levels. By transwell assay and transplanted nude mice, we observed that p120ctn knockdown promoted the invasive ability of lung cancer cells both in vitro and in vivo. Conclusions Our data reveal that ablation of p120ctn enhances the invasion and metastasis probably due to reduce the expression of E-cadherin and β-catenin in lung cancer ceils.
