目的:研究在Poly I:C刺激下原发性干燥综合征(pSS)患者单核细胞分泌Ⅰ型干扰素的能力。方法:分别以50μg/ml和100μg/ml的Poly I:C刺激pSS和健康个体的单核细胞,于刺激后第6和第12小时采用ELISA法检测培养基内IFN-α和IFN-β水平。分别用pSS患者和健康个体的血清培养来自二者的单核细胞,并用50μg/ml的Poly I:C刺激12小时后ELISA法检测培养基内IFN-α和IFN-β水平。结果:Poly I:C能够诱导pSS患者和健康个体的单核细胞呈时间和计量依赖性地释放IFN-α和IFN-β,pSS患者释放IFN-α和IFN-β的能力强于健康个体。pSS血清可以增强Poly I:C诱导单核细胞释放IFN-α和IFN-β的能力。结论:pSS患者单核细胞Poly I:C刺激下分泌Ⅰ型干扰素能力增强,而患者血清可以进一步增强其反应性。天然免疫应答异常可能在pSS发病机制中发挥重要作用。
Objective:To investigate the production of IFN-α and IFN-β by monocytes from primary Sjgren Syndrome(pSS) patients in response to Poly I:C.Methods: The monocytes from pSS patients and healthy controls were treated with 50 μg/ml or 100 μg/ml Poly I:C.The IFN-α and IFN-β level in medium culture was determined by ELISA after stimulated 6 and 12 hours.In the presence or absence of the serum from pSS patients and healthy individuals,we treated the monocytes with 50 μg/ml Poly I:C 12 hours,and then determined the concentration of IFN-α and IFN-β in medium culture.Results:The production of IFN-α and IFN-β by monocytes from pSS patients were induced time and dose dependently by Poly I:C simulation.Increased IFN-α and IFN-β production was observed in monocytes from pSS patients.The serum from pSS patients could enhance the production of IFN-α and IFN-β triggered by Poly I:C.Conclusion: Monocytes from patients with pSS appear more sensitive to Poly I:C,resulting in increased IFN-α and IFN-β production,which could be enhanced by the patients’serum.This abnormality may be a typical characteristic of the pathogenesis of pSS.
