目的检测Cajal间质细胞(Iccs)及突触素(SY)在肛门直肠畸形胎鼠直肠肠壁的胚胎发育过程,探讨其表达的意义。方法应用免疫组化、免疫荧光和Westemblot方法检测正常与肛门直肠畸形胎鼠直肠末端组织c-kit、SY的表达情况。结果正常组结肠及直肠壁内肠壁肌层及黏膜肌层c—kit、SY大量表达;畸形组胎鼠直肠末端壁内的分布均比对照组明显减少,发育延迟,但随着远离盲端,c-kit、SY出现并逐渐增多。正常胎鼠直肠末端壁内c—kit、SY总蛋白量明显高于畸形组(P〈0.05)。结论Cajal间质细胞及突触素的分布异常是肛门直肠畸形动物模型直肠末端壁的重要病理改变之一。
Objective To detect the expressions of interstitial cell of Cajal (ICC)and synaptophysin and their significance in the development of distal rectum in fetal rat model of anorectal malformation( ARM ). Methods The expessions of c-kit and synaptophysin in the distal recturm were detected by method,immunofluorescence,and Western blot in normal and ARM fetal rats. Results In control group, c-kit and synatophysin were highly expressed in the muscular layer of mucosa of the colon and rectum.Compared with control group, the expressions of c-kit and synatophysin were lower and the development was delayed in ARM group.The expressions of c-kit and synatophysin increased from distal rectum to proximal rectum in ARM group.The total levels of c-kit and SY proteins were significantly higher in control group than in ARM group(P 〈 0.05 ). Conclusion Abnormal distribution of ICC and synaptophysin in the distal rectum is one of the most important pathologic changes in ARM fetal rots.
