目的研究穿透支原体脂质相关膜蛋白能否诱导激活小鼠巨噬细胞核因子κB(NF-κB),以了解穿透支原体潜在的致病性。方法用从穿透支原体中提取的脂质相关膜蛋白刺激小鼠巨噬细胞,经Western blot和间接免疫荧光等方法检测NF-κB是否被激活,以及NF-κB抑制剂PDTC对NF-κB活性的影响。结果穿透支原体脂质相关膜蛋白能诱导小鼠巨噬细胞激活NF-κB,且NF-κB抑制荆PDTC能部分抑制穿透支原体脂质相关膜蛋白所诱导的NF-κB激活。结论穿透支原体脂质相关膜蛋白能激活小鼠巨噬细胞NF-κB,因而可能与其致病性有关。
Objective In order to investigate the potential pathogenicity of Mycoplasma penetrans(Mpe), the activation of nuclear factor κB(NF-κB) was studied in mouse macrophages stimulated by lipid - associated membrane proteins (LAMPs) prepared from Mpe. Methods Mouse macrophages were stimulated with Mpe LAMPs to analyze the aetivitation of NF-κB, and the effect of pyrrolidine dithiecarbamate ( PDTC ), an inhibitor of NF-κB, on NF-κB activation was also studied by indirect immunofluorescence and western blot. Results Mpe LAMPs can trigger NF-κB activation in mouse macrophages, and PDTC can partly inhibit the activation of NF-κB. Conclusion Mpe can trigger NF-κB activation in mouse macrophages, thus it may be an important etiological factor.
