本研究旨在研究低氧孵化条件下,藏鸡和低地鸡胚胎肝脏egl-9家族低氧诱导因子1(egl-9family hypoxia-inducible factor 1,EGLN1)基因的表达差异及其与藏鸡胚胎低氧适应的关系。以15%氧浓度下孵化至16 d的藏鸡和白来航鸡胚为研究对象,利用Real-time PCR和western-blot方法对鸡EGLN1基因肝脏组织表达特点进行研究。结果表明:随着氧浓度的降低,白来航鸡胚肝脏中EGLN1基因m RNA表达量极显著下降(P〈0.01);在低氧条件下,白来航鸡胚中EGLN1基因的表达量显著地低于藏鸡胚胎(P〈0.05);白来航鸡中EGLN1基因蛋白表达量随着氧浓度下降而下降,趋势与m RNA表达量一致。而藏鸡EGLN1基因的转录和翻译水平均为受到氧气浓度降低的影响。由此可知,鸡胚通过下调EGLN1基因表达从而应对低氧反应。上述结果为进一步研究EGLN1基因与低氧适应的关系提供了依据。
Recently,researches revealed that eg l-9 family hypoxia-inducible factor 1(EGLN1) gene from the hypoxia-inducible factor(HIF) pathway involved with hypoxia adaptation of Andeas and Tibetans to analysis gene expression of EGLN1 gene of Tibetan Chicken and White Leghorns and its effect on hypoxia adaptation of Tibetan Chicken,the chickens were incubated to days 16 separately in hypoxia and normoxia.Real-time PCR,western blotting techniques were used determinate the expression of EGLN1 gene of embryo liver.The expression of livers with White Leghorn of EGLN1 gene in the normoxia condition was significantly higher in value than that of hypoxia condition(P〈0.01).Oxygen concentration of 15% had a significant different effect on the expression of livers between the White Leghorn and Tibetan Chicken(P 〈0.05).The results showed that the protein expression of EGLN1 gene in the liver was declined with the decreasing of oxygen concentration.In hypoxia,expression of EGLN1 gene in white leghorn is lower than that of Tibetan chicken,and the trend was similar with the expression of the m RNA in liver.Accordingly,to response the stress of hypoxia,the chicken decreased the expression of EGLN1 gene.This work provides reference for future studies on the association of the EGLN1 gene expression with the adaptation to hypoxia.
