The optimum in vitro shoot section explants growing medium for Prunus mume cultivar Tiegu Hong was the modified Quoirin and Lepoivre medium supplemented with Quoirin and Lepoivre medium large elements, Perez medium trace elements, Murashige and Skoog medium (MS) organic compounds, and double MS Fe salt. This modified medium was used to resolve a serious of problems faced in Tiegu Hong tissue culture such as seedling chlorosis and shoot tip death. The optimum proliferation medium for Tiegu Hong was the im- proved Quoirin and Lepoivre medium supplemented with 1.0 mg·L^-1 6-BA, 0. 05 mg·L^-1 NAA and 4 mg·L^-1 AgNO3. Attention should be paid to the application of AgNO3 because of long period of use may lead to the plantlets malformation. The optimum rooting medium for Tiegu Hong was the improved 1/2 Quoirin and Lepoivre medium supplemented with 0. 3mg·L^-1 NAA and 0. 1 mg·L^-1 IBA.
