目的:观察糖皮质激素性骨质疏松症(GIOP)大鼠骨、肾组织同源异形盒基因5(Dlx5 mRNA)及蛋白表达,探讨GIOP的发病机制、补肾中药复方的疗效机制及其与健脾、活血中药复方及骨疏康颗粒的比较。方法:肌肉注射地塞米松复制GIOP大鼠模型,实验设正常组、模型组、补肾组、健脾组、活血组、阳性对照组。造模及灌胃给药9周。测定股骨骨密度,RT-PCR及Western Blot检测骨、肾组织Dlx5 mRNA及蛋白表达。结果:与正常组比较,模型组股骨骨密度明显降低(P〈0.01)、骨组织Dlx5 mRNA及蛋白表达明显降低(P〈0.01)、肾组织Dlx5 mRNA及蛋白表达明显升高(P〈0.01);与模型组比较,补肾组及阳性对照组股骨骨密度明显升高(P〈0.01)、骨组织Dlx5 mRNA及蛋白表达明显上调(P〈0.01)、肾组织Dlx5 mRNA及蛋白表达明显下调(P〈0.01)。结论:骨组织Dlx5 mRNA及蛋白表达降低、肾组织Dlx5 mRNA及蛋白表达升高可能是GIOP的发病机制之一;补肾中药复方可能通过上调骨组织Dlx5 mRNA及蛋白表达、下调肾组织Dlx5 mRNA及蛋白表达,有效防治GIOP。
Objective: To observe the mRNA and protein expression of Dlx5 in bone and kidney tissues of the rats with glucocorticoid-induced osteoporosis(GIOP),discuss the pathogenesis of GIOP and the effective mechanism of reinforcing the kidney herbs compound and compare the therapeutic effect and adjustable function of it with strengthening the spleen herbs compound and promoting blood circulation herbs compound.Methods: We established the GIOP rat models by intramuscular injection of dexamethasone.We divided all the rats into normal group,model group,reinforcing the kidney group,strengthening the spleen group,promoting blood circulation group and positive control group.After 9 weeks of establishing models and administrating by gavage,we detected femur bone mineral density by using the XR-26 dual energy X-ray absorptiometry.We detected mRNA expression of Dlx5 in bone and kidney tissues by Real-Time Quantitative RT-PCR and detected their protein expression by Western Blot.Results: Compared with normal group,femur bone mineral density of model group decreased significantly(P0.01),mRNA and protein expression of Dlx5 decreased significantly(P0.01) in bone tissues and increased significantly(P0.01) in kidney tissues.Compared with model group,femur bone mineral density of reinforcing the kidney group increased significantly(P0.01),mRNA and protein expression of Dlx5 up-regulated significantly(P0.01) in bone tissues and down-regulated significantly(P0.01) in kidney tissues.Conclusion: Maybe it is one aspect of the pathogenesis of GIOP that mRNA and protein expression of Dlx5 decreases in bone tissues and increases in kidney tissues;reinforcing the kidney herbs compound has more evident preventative and therapeutic effect on GIOP by up-regulating mRNA and protein expression of Dlx5 in bone tissues and down-regulating its expression in kidney tissues than strengthening the spleen herbs compound and promoting blood circulation herbs compound.
