目的:高产透明质酸菌种的选育是提高透明质酸产量和质量的关键。方法:以马疫链球菌SH~0为出发菌,对其进行紫外诱变和NTG诱变选育。结果:筛选出溶血素和透明质酸酶双缺陷型突变菌株FJ-23,其透明质酸产量提高了7倍;突变株经6次传代,其产酸量及HA的相对分子量保持稳定,为今后进一步研究发酵法生产透明质酸打下基础。
It was very critical for improving the quality and quantity of HA to select a high hyaluronic acid - producing stain. A selected mutant, designated as Streptococcus equi FJ - 23 was selected from wild - type Streptococcus equi SH - 0 by UV and NTG inducing mutagenesis, which did not produce streptolysin or hyaluronidase. The mutant could produce HA seven times as much as the original strain. Hyaluronic acid productivity and its relative molecular weight of the mutant kept stable after 6 generations of reproduction, which displayed perfect genetic stability. The research laid a good foundation for the further study on producing HA by fermentation.
