目的:观察降糖通脉片对四氯化碳所致小鼠急性肝损伤的修复作用并探讨其可能的作用机制。方法:将40只昆明种小鼠按随机数字表法分成空白对照组、模型组、降糖通脉片组、阳性对照组(水飞蓟宾胶囊)4组各10只。预防性灌胃给药5 d后,通过腹腔注射0.2%CCl4花生油溶液,建立小鼠急性肝损伤模型,测定小鼠血清中ALT、AST、TG、TBIL、LDH和ALP水平及肝组织中MDA含量和SOD活力,计算肝脏指数并对肝组织进行病理学检查。结果:降糖通脉片组能降低CCl4导致的急性肝损伤小鼠肝脏指数和血清中ALT、AST等指标水平,降低肝脏组织中MDA的含量,增强SOD的活力。通过病理学检测,降糖通脉片组能明显改善肝脏组织的病理变化。结论:降糖通脉片对四氯化碳引起的小鼠急性肝损伤具有显著的修复作用,其作用机制可能与清除体内自由基和抗氧化应激有关。
Objective To evaluate the protective effect of Jiang Tang Tong Mai Tablet on carbon tetrachloride (CCL4 ) induced acute liver injury in mice and its possible mechanism as well. Method: The 40 mice were randomly divided into normal control, model group, JT group, then there is silibinin group( n = 10). The mouse model of acute liver injury was established by injecting 0. 2% CCl4 , after five days preventive administration. The serum levels of ALT, AST, TBIL, TG, LDH and ALP were determined, and the mice were killed to calculate the index of liver. MDA and SOD were detected in the liver homogenate. Histopathological changes of the liver were observed by HE and Oil red staining. Result JT reduce the liver index in mice insignificantly, decreased the level of ALT, AST, TBIL, TG, LDH and ALP in serum and the content of MDA in the liver homogenate, furthermore increased the activity of SOD in liver tissue. Conclusion:JT could protect the acute liver damage induced by CCL4 and its mechanism may be related to the elimination of free radicals and anti-oxidative stress.
