目的探讨C型凝集素16A(CLEC16A)在中枢神经系统(CNS)炎症中的表达变化及细胞定位。方法应用SD大鼠侧脑室内注射脂多糖(LPS)建立脑炎症模型,利用PCR及Western blot检测术后大脑皮层中CLEC16A在mRNA水平及蛋白水平的表达变化;免疫荧光检测CLEC16A在侧脑室内注射LPS后的定位、细胞分布及细胞的状态。结果在大鼠脑炎症模型中CLEC16A mRNA和蛋白水平均有明显变化,分别在注射后9或12 h开始增加,并一直维持在较高水平。免疫荧光双标发现表达较高CLEC16A主要定位与大鼠脑中的星形胶质细胞及神经元中。同时检测脑中星形胶质细胞活化的情况发现表达增多的CLEC16A主要定位于增殖的星形胶质细胞。结论CLEC16A可能参与星形胶质细胞的增殖,在神经系统炎症反应过程中发挥着重要的作用。
Objective To observe the expression and cellular localization of C-type lectin 16A(CLEC16A) in the central nervous systerm(CNS) inflammation and investigate its function in the process of CNS inflammation.Methods The model of brain inflammation was prepared in SD rats by the intracerebroventricular injection of LPS.The expression of CLEC16A mRNA and protein were assayed by PCR and Western blot.The distribution,localization and cellular state of CLEC16A in cerebral cortex were investigated by immunofluorescence staining.Results After LPS intracerebroventricular injection,the expression of CLEC16A mRNA and protein were increased in time-dependent way in a cerebral corter,which was increased 9 or 12 hours after injection,and maintained in a high level.Double immunofluorescence staining indicated that up-regulated CLEC16A was mostly expressed in astrocytes and neurons,and change of its expression was mainly observed in astrocytes and neurons.In further,astrocytes were colocalization with PCNA(proliferating cell nuclear antigen) after LPS injection.Conclusion CLEC16A may play an important role in CNS inflammation disease,and it may participate in mediating astrocyte proliferation during CNS inflammation.
